Gene cloning is a fundamental technique in molecular biology that enables the isolation, amplification, and manipulation of specific DNA sequences for research, biotechnology, and therapeutic applications. The cloning process involves a series of interconnected steps, including preparation of the DNA insert, vector preparation, ligation, transformation into suitable host cells, screening of recombinant clones, and propagation of validated constructs. Restriction enzymes play a pivotal role by generating precise DNA fragments with compatible ends, while DNA ligase facilitates the formation of stable recombinant DNA molecules. Various cloning vectors, such as pBR322 and pUC plasmids, provide essential features including origins of replication, selectable markers, and multiple cloning sites for efficient gene insertion and maintenance. Modern cloning approaches, including T/A cloning, Cre/LoxP recombination, and Gateway (att) recombination, have simplified gene transfer and expanded experimental flexibility. Selection of an appropriate host system is equally important and depends on the intended application. Prokaryotic hosts such as Escherichia coli offer rapid growth and cost-effective protein production, whereas eukaryotic systems, including yeasts and mammalian cell lines, enable complex post-translational modifications required for functional therapeutic proteins. Together, advances in restriction enzymes, cloning vectors, ligation strategies, and host expression systems have established recombinant DNA technology as a cornerstone of modern biotechnology, supporting applications in gene therapy, vaccine development, functional genomics, and recombinant protein production
